QFILL2 Protocol for Deep Well Boxes

1.)   Fill autoclaved bottle with correct media (including antibiotic).

2.)   Attach silicon tube from back of QFILL2 to short metal tube on bottle. Do the same with the silicon tube attached to the maniflold, but connect to the long, bent tube on the bottle.  Please make sure you have chosen the proper manifold and the correct tubing for the antibiotic that you are using.

 

3.)   Tighten the bottle lid and squeeze manifold tube under black tab in middle of QFILL2.

 

4.)   Turn power switch in the back to the ON position.

5.)   Reset QFILL2 and verify the correct volume (e.g. 250 or 1200ml) and plate type (96).

·      Press the “Vol” button.  If the volume you need is not on the list, hit the “Menu” button twice. 

·      The first option will be “Change Volumes” and use the up/down arrow buttons to make sure the asterik is in front of “Change Volumes”.  Press the “Enter” button.

·      Select one of the volumes to change by moving the asterik  with the up/down arrow buttons.  Press “Enter” button when the volume you want to change is selected

·      Now the volume will say “Edit” after the number and the first digit will be underlined.  Use the up/down arrow button to change the number for that digit and then press “Enter” and the next digit will be underlined for editing. 

·      When all the digits have been changed to give the desired volume, press “Enter” once more and the “Edit” prompt after the volume number will disappear.  Then press “Menu” twice and you will be back to the start screen.

 

6.)    To Refill Bottle:

·      1st break vacuum seal by turning bottle cap.

·      Unclamp manifold tube.

·      Fill bottle with media.

·      Clip manifold tube again.

 

7.)   Purge air out of manifold tube and make sure media streams out of every prong (do not waste too much media).

·      Press the “Purge” button to get to the purge screen

·      Press the “Purge” button again and hold it down until the media starts to flow out of the manifold.

·      Press the “Start/Stop” button to get back to the main menu.

 

8.)   Fill boxes by pressing “Start/Stop” button.

 

 

9.)   Cleaning Instructions: Purge with ~500ml each of water, 70% ethanol, then water again.  Autoclave the manifold (remove the manifold plugs), bottle, and bottle top together.
